Saussurea lappa C.B. Clarke (Compositae) is indigenous to India and Pakistan. The dried root of S. lappa has been traditionally used for alleviating pain in abdominal distention and tenesmus, indigestion with anorexia, dysentery, nausea, and vomiting. Santamarin is a sesquiterpene lactone isolated from S. lappa. In the present study, santamarin inhibited inducible nitric oxide synthase (iNOS) protein, reduced iNOS-derived nitric oxide (NO), suppressed COX-2 protein and reduced COX-derived PGE(2) production in lipopolysaccharide (LPS)-stimulated RAW264.7 and murine peritoneal macroges. Similarly, santamarin reduced tumor necrosis factor-&al; (TNF-&al;) and interleukin-1β (IL-1β) production. In addition, santamarin suppressed the srylation and degradation of IκB-&al; as well as the nuclear translocation of p65 in response to LPS in RAW264.7 . Furthermore, santamarin induced heme oxygenase (HO)-1 expression mRNA and protein level that plays a cytoprotective role against inflammation. The induction of HO-1 is primarily regulated at the transcriptional level, and its induction by various agents is mediated by the nuclear transcription factor E2-related factor 2 (Nrf2), master regulator of antioxidant responses. Unbound Nrf2 translocates into the nucleus and binds to the antioxidant response the upstream promoter region of many antioxidative genes, where it initiates their transcription. The effects of santamarin on LPS-induced NO, PGE(2), TNF-&al;, and IL-1β production were partially reversed by the HO-1 inhibitor, tin protoporrin (SnPP). Therefore, our data suggest that the anti-inflammatory effect of santamarin in macroges may be exerted through a novel mechanism that involves HO-1 expression.

Santamarine and 9beta-acede inhibited L1210 growth, colony formation and [(3)H]-thymidine incorporation in time- and concentration-dependent manners. Flow cytometry studies showed that Santamarine and 9beta-acede blocked L1210 in the G(2)/M se of the cycle. DAPI staining and caspase activity assays showed Santamarine and 9beta-acede induced apoptosis and activated caspase 3 in L1210 . CONCLUSIONS: These results indicated that Santamarine, 9beta-acede and 9beta-aceide exhibit significant anticancer activities in vitro. The inhibitory effects of Santamarine and 9beta-acede on L1210 are cytothan just cytostatic. They block mitosis and reduce uptake of thymidine. The mechanism of the cytoantamarine and 9beta-acede to L1210 could be related to alkylation of the sulfhydryl enzymes involved in nucleic acids and protein synthesis, as previously found for other sesquiterpenes with the al-methylene-gamma-lactone moiety present in Santamarine, 9beta-acede and 9beta-aceide. It may also be related to suppression of microtubular proteins. Santamarine and 9beta-acede induced apoptosis of L1210 via activation of caspase 3.
